A Phospholipids B test kit for the quantitative determination of phospholipid content of serum and plasma is available for use on most automated chemistry analyzers.
This assay utilizes an in vitro enzymatic colorimetric methodology in a simple one step, direct determination, with high specificity. It is readily automatable.
Phospholipids in serum or plasma (lecithin, lysolecithin and sphingomyelin) are hydrolyzed to free choline by phospholipase D. In the presence of choline oxidase and peroxidase, the free choline is then estimated colorimetricaly, by taking advantage of the color reaction with phenol and 4-aminoantipyrine at 505 nm.
The within-run precision of this assy is 1.3%CV, 1.4%CV and 1.4%CV for mean concentrations of 132mg/dL, 228mg/dL and 324mg/dL, respectively. The linear range of the assay is 0-1000mg/dL. There is no significant interference from bilirubin, hemoglobin, glucose, ascorbic acid, uric acid or anticoagulants such as sodium citrate, heparin, ammonium oxalate or EDTA.
