Capable of providing DNA yields as high as 100%, the UltraClean 15 DNA-purification kit utilizes uniform silica-binding beads to purify DNA fragments from TAE or TBE agarose gels. Purification requires 10 minutes for reaction solutions and 15 minutes for agarose gels. The process requires only one wash and elution step, removing ethidium bromide, linkers, and labels.
The kits silica-binding technology can be used in a batch format to allow the purification of small, medium, or large amounts of DNA, and utilizes a simple-to-follow protocol. After electrophoresis, the desired DNA band is cut from an agarose gel and melted in a high-polarity chaotropic salt solution. The DNA binds to the UltraBIND silica particles in the presence of UltraSALT and is then pelleted in a microcentrifuge. After centrifuging, the melted agarose is discarded, the pellet is washed, and the concentrated DNA is collected in Tris buffer or water.
