A reagent developed for the efficient transfer of functional proteins and antibodies to the cytoplasm of living cells is available from Polyplus-transfection. PULSin Protein and Antibody Delivery Reagent enables researchers to study lethal proteins and control the level and time course of proteins in cells. It also permits the delivery of blocking antibodies in addition to the targeting of intracellular proteins in living cells using antibodies without fixation. PULSin incorporates a proprietary cationic lipid that noncovalently complexes with many proteins and antibodies. These complexes are internalized via anionic cell-adhesion receptors present on all cells and released into the cytoplasm, where they disassemble. The process is general, nontoxic, and delivers unmodified, functional proteins to the cytoplasm with up to 98% efficiency.
